Health bosses are hoping to have vaccinated more than 2,000 people at the new site in the Maldwyn Leisure Centre by the end of the weekend.
More than 12,000 people have now been vaccinated throughout Powys.
Powys Teaching Health Board relocated the Covid-19 vaccination centre from the Park Street Day Centre in order to cope with larger numbers. It undertook 500 vaccinations within its first day.
The Newtown site is part of the national network of vaccination centres, allowing eligible people to book vaccinations online.
Newtown, Montgomery and Llanidloes GP surgeries will be joining the vaccination programme from Monday.
Anyone who is eligible will have received a letter from the health board and should book an appointment through the online system and not call their GP.
